What is NAD+ (nicotinamide adenine dinucleotide)?

NAD+ is a coenzyme found in every living cell in our body and is essential for maintaining cellular health and energy.

NAD+ is an electron shuttle service and participates in redox reactions by accepting and donating electrons. In its oxidised form it exists as NAD+, the form that takes up electrons and in its reduced form, it exists as NADH which donates its electrons to molecules in the body. This electron exchange is essential for life giving chemical processes and reactions occurring in the cells. Key Functions of NAD+ include:

How is NAD+ made?

NAD+ is primarily produced through the salvage pathway, recycling existing NAD+ precursors such as niacinamide (NAM) and nicotinamide riboside (NR) instead of creating it from scratch. NR and NAM can then be converted to NMN (nicotinamide mononucleotide) under the influence of the enzyme nicotinamide phosphoribosyl transferase (NAMPT).

The enzyme nicotinamide mononucleotide adenylyl transferase (NMNAT aka NMN transferase) then steps in and converts this NMN to NAD+.

The body prioritises recycling existing NAD+ precursors from NR and niacinamide (NAM) via the salvage pathway as this is more energy efficient for the cells than making NAD+ from scratch. The salvage pathway also has the advantage of using the NAMPT enzyme to adjust NAD+ levels for the cells, based on the needs of the cell.
